Engineering extracellular vesicles with macrophage membrane fusion for ameliorating imiquimod-induced psoriatic skin inflammation

Introduction Herein, we developed an engineered extracellular vehicle (EV)-based method for ameliorating inflammatory responses in psoriasis.Methods EVs, derived from annexin A1 (ANXA1) overexpressing T cells, were co-extruded with M2 macrophage membrane to obtain EVs. In vitro, the effect of EVs on polarization was evaluated by real-time PCR. imiquimod (IMQ)-induced psoriasis-like mouse model, efficacy psoriatic inflammation Psoriasis Area and Severity Index (PASI) score immunohistochemistry staining after subcutaneous injection EVs.Results The not only preserved high stability but also retained reprogramming potential ANXA1 overexpressed cells. successfully reduced PASI levels pro-inflammatory cytokines, including IL-1β, IL-6, TNF-α. Along biosafety, administration rescued histomorphological changes spleen, liver, kidney.Conclusions exhibited alleviate psoriasis, providing new insights strategies immunotherapies psoriasis.
